操作臺(tái)的滅菌處理:
1)無(wú)菌室及無(wú)菌操作臺(tái)(laminarflow)用紫外燈照射30-60分鐘滅菌,70%ethanol擦拭無(wú)菌操作抬面,并開(kāi)啟無(wú)菌操作臺(tái)風(fēng)扇運(yùn)轉(zhuǎn)10分鐘。
2)細(xì)胞用的培養(yǎng)基如有相同切記不可共享培養(yǎng)基,以避免失誤混淆或細(xì)胞間污染。 (注:實(shí)驗(yàn)操作應(yīng)在抬面之*無(wú)菌區(qū)域,勿在邊緣之非無(wú)菌區(qū)域操作。)
培養(yǎng)細(xì)胞將復(fù)合細(xì)胞計(jì)數(shù)要求的細(xì)胞懸液分裝入培養(yǎng)瓶?jī)?nèi),將培養(yǎng)瓶放入37℃和5%CO2的培養(yǎng)箱內(nèi)2-4小時(shí)(或者24-48小時(shí))后換液繼續(xù)培養(yǎng)培養(yǎng),換液的時(shí)間由細(xì)胞情況而定。
蘇因嚴(yán)格遵守“快速融化”原則:必須將凍存在-196℃液氮中的細(xì)胞快速融化至37℃,使細(xì)胞外凍存時(shí)的冰晶迅速融化,避免冰晶緩慢融化時(shí)進(jìn)入細(xì)胞形成再結(jié)晶,對(duì)細(xì)胞造成損害。
