S100A14/15抗體 的步驟：
1、制備抗原。
2、選擇實(shí)驗(yàn)動(dòng)物。
3、動(dòng)物免疫。
4、試取血進(jìn)行測試，看看是否成功免疫。
5、如果成功免疫，殺死實(shí)驗(yàn)動(dòng)物，采集全部血清。
6、純化出抗體。
7、鑒定抗體。包括純度以及特異性。
儲存溫度：存在于血清、組織培養(yǎng)上清液或腹水中的抗體在-20℃下能夠*儲存。低溫存放不會損傷抗體活性。抗體的工作液通常儲存于4℃中。此溫度下，抗體的活性能穩(wěn)定數(shù)月至數(shù)年亦不損失。抗體溶液不應(yīng)反復(fù)凍融，因?yàn)榉磸?fù)凍融將導(dǎo)致部分抗體失活并產(chǎn)生不需要的蛋白—蛋白凝聚物。抗體的凝聚能導(dǎo)致活性喪失。這是由于抗原結(jié)合位點(diǎn)的空間構(gòu)象被破壞，或由于產(chǎn)生的不溶物被離心或過濾而除去。冷凝蛋白類的抗體不能存放于4℃。因?yàn)樵?/span>4℃下此類抗體會發(fā)生沉淀。一些鼠源性IgG3亞類的抗體有此特性。如果抗體在4℃下發(fā)生沉淀，就應(yīng)該存放于室溫下的疊氮鈉溶液中。
